MAPseq or Multiplexed Analysis of Projections by Sequencing is a RNA-Seq based method for high-throughput mapping of neuronal projections. It was developed by Anthony Zador and his team at Cold Spring Harbor Laboratory and published in Neuron, a Cell Press magazine. It has been cited over 300 times. The method works by uniquely labeling neurons in a source region by injecting a viral library encoding a diverse collection of RNA sequences ("barcodes"). The barcode mRNA is expressed at high levels and transported into the axon terminals at distal target projection regions. Following this, the cells from source and putative target regions of interest are harvested, and their RNA is extracted and sequenced. The fragment molecular orbital method (FMO) was developed by Kazuo Kitaura and coworkers in 1999. FMO is deeply interconnected with the energy decomposition analysis (EDA) by Kazuo Kitaura and Keiji Morokuma, developed in 1976. The Bergmann degradation is a series of chemical reactions designed to remove a single amino acid from the carboxylic acid (C-terminal) end of a peptide. First demonstrated by Max Bergmann in 1934, it is a rarely used method for sequencing peptides. The later developed Edman degradation is an improvement upon the Bergmann degradation, instead cleaving the N-terminal amino acid of peptides to produce a hydantoin containing the desired amino acid. The Bergmann degradation follows the earlier work of Bergmann and his close colleague Leonidas Zervas, combining the organic azide degradation of the Curtius rearrangement with the Bergmann-Zervas carbobenzoxy method, which they designed to occur under relatively mild conditions so as to allow peptide sequencing. Several phase 1 clinical trials have been performed by experts in the field of adoptive immunotherapy of cancer. Hans Klingemann and Sally Arai completed a US trial at Rush University Medical Center (Chicago) in renal cell cancer and melanoma patients in 2008, and Torsten Tonn, MD and Oliver Ottmann, MD completed the European trial at the University of Frankfurt in patients with various solid and hematological malignancies in 2013. Armand Keating at Princess Margaret Hospital in Toronto conducted a trial in which NK-92 cells were given to patients who had relapsed after autologous bone marrow transplants for leukemia or lymphoma. In all clinical trials so far, NK-92 cells were administered as a simple intravenous infusion, dosed two or three times per treatment course, and given in the outpatient setting. Of the 39 patients enrolled across the three studies, 2 serious (grade 3–4) side-effects occurred during or after the infusion of NK-92 cells, the side effects disappeared afterward. The doses given to patients ranged from 1 × 108 cells/m2 to 1 × 1010 cells/m2 per infusion. Patients received between two and three infusions over a period of less than a week. About one-third of the treated patients had clinically meaningful responses with some of them fully recovering.
